台法攜手研發 高頻音波治腦疾病
腦中風是現代常見的疾病，中風後的治療與復健對於家人來說，更是漫長又花錢，一項由科技部與法國國家研究署（ANR）共同補助的國際合作計畫，歷經三年研究成功發現，利用高頻音波可於短暫時間內打開血腦屏障，使藥物進入中樞神經系統，對中風、失智症等腦部疾病達到更好的治療效果。
科技部昨日召開記者會發表這項研究成果，政務次長謝達斌表示，台法雙方的研究團隊，我方由台大醫院醫師陳文翔擔任計畫主持人，法方則由里昂第一大學副教授Claude INSERRA擔任，雙方共同合作率領研究團隊已發表多篇學術論文，研究成果也在今年二月獲刊於英國科學期刊《Scientific Reports》。
擅長超音波研究、治療的陳文翔醫師表示，人類大腦內存在一特殊結構稱為血腦屏障（Blood Brain Barrier，BBB），由於此特殊結構的存在，外在有害物質不易進入腦部破壞腦組織，但若腦部發生病變，例如感染或出現腫瘤，九八％的藥物難以進入腦部，造成中樞神經相關疾病治療上的窒礙。
陳文翔指出，近年多方研究皆證實高頻音波，如超音波所觸發的空穴效應（Cavitation Effect），可於短暫時間內，約○‧五至四小時，有效增強血腦屏障部位的通透性，提供藥物得以進入中樞神經系統的治療窗口。
台法研究團隊已初步取得多項成果，包括能即時監控、回授系統，有效持續穩定並增強血腦屏障開啟效果，以及首度發現超音波觸發血腦屏障開啟的分生機制，可能與腦內血管細胞膜上的TRPV4通道有關。
研究也發現，利用另一種高能量音波系統，即震波，可更簡單開啟血腦屏障，讓原本無法穿越血腦障壁的藥物，大量穿入腦部，包括大分子基因，達到基因轉殖效果，不只有助於治療腦中風，也可應用在治療失智及腦瘤研究。
陳文翔說，目前該研究團隊正進行腦部膠質母細胞瘤動物治療模組的建立，期望利用新發展的血腦屏障開啟技術，增加穿入腦部藥物濃度，以增強藥物對惡性膠質母細胞瘤的治療效果。未來進一步探討、開發針對中樞神經系統與腦部相關的基因、藥物療法，解決習用療法因血腦屏障所造成的困境。
